ABSTRACT

OBJECTIVE@#This study aimed to investigate the effect of calcium ion (Ca²⁺) on the migration and osteogenic differentiation of human osteoblasts and explore the proper concentration and correlation mechanism.@*METHODS@#A series of Ca²⁺ solutions with different concentrations was prepared. Osteoblast migration was assessed by Transwell assay, and proliferation was studied via the CCK-8 colorimetric assay. The mRNA expression of osteogenic genes was examined via reverse transcription-polymerase chain reaction (RT-PCR), and the mineralized nodule was examined by alizarin red-S method. After calcium sensitive receptor (CaSR) antagonism, Ca²⁺-induced migration and osteogenic differentiation were analyzed.@*RESULTS@#In the migration experiment, 2, 4, and 6 mmol·L⁻¹ Ca²⁺ could promoted osteoblast migration at three timepoints (8, 16, and 24 h), whereas 10 mmol·L⁻¹ Ca²⁺ considerably inhibited migration at 8 h. The Ca²⁺ concentration range of 2-10 mmol·L⁻¹ could promote proliferation, osteogenic differentiation, and mineralization of human osteoblasts. Moreover, mineralization was predominantly induced by 8 and 10 mmol·L⁻¹ Ca²⁺. CaSR antagonism could reduce Ca²⁺-induced migration and osteogenic differentiation of human osteoblasts.@*CONCLUSIONS@#Low Ca²⁺ concentration favored osteoblast migration, whereas high Ca²⁺ concentration favored osteogenic differentiation. The Ca²⁺ concentrations of 4 and 6 mmol·L⁻¹ could substantially induce osteoblast migration and osteogenic differentiation, and the Ca²⁺-CaSR pathway participated in signal transduction.

ABSTRACT

Objective To study the clinical efficacy of low dose thyroxine combined with irbesartan hydrochlorothiazide in the treatment of elderly patients with severe heart failure. Methods 60 cases of elderly patients with severe heart failure who were admitted from January 2015 to December 2016 were selected as the research object in this study. They were randomly divided into control group and experimental group, each group was divided into two groups, each with a total of 30 cases. Patients in the control group were treated with conventional therapy, while the experimental group was treated with low dose thyroxine combined with irbesartan and hydrochlorothiazide. Comparative analysis of clinical efficacy of the experimental group and control group. Results After treatment, the total effective number of patients in the experimental group was 26 cases, the effective rate was 86.7%. The total effective number of patients in the control group was 21 cases, the effective rate was 70.0%. Therefore, the effective rate of the experimental group was significantly higher than that of the control group, with statistical difference (P<0.05). After treatment, the experimental group of patients with left ventricular ejection fraction index, four triiodothyronine and three triiodothyronine levels were significantly higher than those before treatment, the control group of patients with left ventricular ejection fraction were significantly higher than before treatment, with statistical difference (P<0.05). Conclusion Low dose thyroxine combined with irbesartan hydrochlorothiazide in the treatment of elderly patients with severe heart failure has better clinical effect, can improve the therapeutic effect in a certain extent, with further clinical promotion and application significance.intervention in two groups. Results The clinical intervention effect of the observation group was better than that of the control group, the stability of the tumor focus, the degree of pain, the quality of life and other aspects were better than the control group, the difference between the two groups was statistically significant (P<0.05). Conclusion Patients with advanced non-smal cell lung cancer chemotherapy choose psychological intervention compound sophora injection combined treatment effect is remarkable, which can effectively improve the patient's clinical symptoms and quality of life.

ABSTRACT

BACKGROUND:Previous researches have focused on the effect of phosphorus compounds on stem cels from animals or from human. But there is no study on the effect of phosphorus ions on human bone marrow mesenchymal stem cels under three-dimensional culture. OBJECTIVE:To explore the effect of phosphorus ions on human bone marrow mesenchymal stem cels under three-dimensional culture. METHODS:There were six groups in the experiment. Human bone marrow mesenchymal stem cels were inoculated in three-dimensional polystyrene scaffolds and then subjected to serum-free growth medium (group 3-GM) or serum-free growth medium containing 4 mmol/L (group 3-4P), 8 mmol/L (group 3-8P) phosphorus ions for 21 days, respectively. Cels cultured on the two-dimensional polystyrene scaffolds were used as control groups (groups 2-GM, 2-4P, 2-8P). Celular proliferation was examined by cel counting kit-8; the mRNA expressions of osteogenic marker genes were assessed by RT-PCR; the formation of mineralized nodules for the osteogenic differentiation of human bone marrow mesenchymal stem cels was examined by Alizarin red staining. RESULTS AND CONCLUSION:Compared with the two-dimensional culture, the growth of human bone marrow mesenchymal stem cels induced by phosphorus ions were more obvious in the three-dimensional polystyrenes scaffolds at days 4, 7, 14 and 21 (P < 0.05). Compared with the group 3-GM, the mRNA expression of colagen type I in groups 3-4P and 3-8P increased more significantly at days 7 and 14 (P < 0.05); the mRNA expression of alkaline phosphatase in groups 3-4P and 3-8P increased more significantly at day 14 (P < 0.05); the mRNA expression of osteocalcin in groups 3-4P and 3-8P increased more significantly at days 14 and 21 (P < 0.05). Mineralized nodules were formed in groups 3-4P and 3-8P but not in group 3-GM at day 21. So we concluded that phosphorus ions can promote proliferation and osteogenic differentiation of human bone marrow mesenchymal stem cels in three-dimensional polystyrenes scaffolds. Compared with the two-dimensional cel culture, the promoting growth effect of phosphorus ions on human bone marrow mesenchymal stem cels in three dimensional polystyrenes scaffolds are more obvious.
